If you already submitted your return, then you will have to wait until your return has been accepted by the IRS and the amended forms are available then you can Amend Your Return .  You cannot simply submit another tax return, as the IRS will reject it for duplicate use of a social security number.

If your return is rejected, then you can just make the corrections and fix the problem.

The 1040X forms are expected to be available on February 23rd.

Why do you want or need to file Married Filing Separate?  Did you file a Joint return or Single?  Joint is usually the best way to file.
