I've already filed, and it's in process, but I just saw this year the personal exemption elimination and max tax deduction of $5K for Married filing separately make me better off taking the standard deduction for the first time in 20 years. Can I amend my return, once it's processed and take the standard deduction as long as my husband also agrees to that on his return?

You will have to wait until the return is either accepted or rejected by the IRS.
If the return is rejected by the IRS you can change to the standard deduction and refile your return.
If the return was accepted by the IRS then you will have to file an amended tax return using Form 1040X to change to the standard deduction.
