I was self-employed Jan - May 2020. As of May I am on the payroll at a company and will have my tax withheld. I will no longer be working for myself in any way. So, is there a way to pay the estimated tax I owe for 2020 on what I earned in the first quarter in one lump sum instead of having to make four separate payments throughout the year? If I can pay in one instalment, do I need to inform the IRS that I am no longer self-employed so they don't chase me for the three remaining quarterly 1040-ES payments? I have TurboTax Self Employed - I assume I can calculate what I owe using that?

You can make a payment at any time. The IRS has actually delayed the requirement to make the 4/15/20 payment until 7/15/20 so you don't need to make a payment until then. You don't need to do anything with the IRS except make the one payment. Yes just pay the full amount on the first quarter. It has to be paid when you earn it anyway, you can delay the 1st & 2nd quarterly estimates until July 15. You don't have to tell the IRS you stopped. Estimates are optional to pay and the IRS won't be expecting them. They don't get sent with your return.
You can just cross out the amount on the 1040ES and write in the amount you are paying. Or pay directly online at https://www.irs.gov/payments
