I just received a letter from NYS. They indicate I owe more than turbo tax said.
I loaded my wife's and my W2s directly from our employers and had no other income. We took the standard deductions. I just updated turbo tax again and it continues to indicate the original amount owed
How do I open a help ticket for this?

TurboTax has a special phone number for help with IRS/state tax letters, which will be easier than going through regular Customer Support. At the page below, click on the blue button that says "Get Help from TurboTax Support". Then it will ask you what tax year is your letter. Then it will ask you what the letter concerns. Then it will show you the phone number during posted business hours.

Turbo tax says there is a calculation guarantee.
I know Turbo tax does not receive anything. How do I go forward claiming the guarantee? Your response was mostly useless as it pertained to Federal IRS not State Taxation. Reading that page, it simply says to "Pay". There is no guidance in checking the calculations
Again, I downloaded directly from my employers into Turbo Tax correctly, It spit out the wrong number.
Did the State say the W-2 amounts were wrong, or that you perhaps failed to claim some other income? What exactly did NYS change? It is highly unlikely that TT's calculations would be wrong this late in the tax season.
Regardless of whether a taxpayer uses software or prepares their return manually, they need to have some basic understanding of what is being sent to the respective governmental entity. Court cases have held that it is ultimately the taxpayers responsibility for their tax return calculations.
Have you looked at your NYS tax return as prepared by TT and compared it to what NYS sent you to see where or why there might be a difference?
If based on your facts all you had was a W-2 and withholding, then this should be a fairly straight forward check on your part. Review your tax return, and compare the state tax to the NYS tax schedules. Does this agree with what TT computed?
It is not uncommon for a notice to be incorrect. I recommend narrowing down where the issue might be and if you are finding that TT did in fact have a calculation error, see the link below:
It's highly unlikely that this is a calculation error in TurboTax. Most likely either some information was entered incorrectly, a question was answered incorrectly, or something was omitted.
What exactly does the letter from the state say was wrong in your tax return? If the letter does not say specifically what they think is wrong, you have to contact them to find out.
Check all of the W-2 information in TurboTax to see if it accurately matches the printed W-2s that you and your wife got from your employers. Sometimes errors occur in importing a W-2. Also make sure that each W-2 was entered for the correct person. When you import a W-2 in a joint return, you have to indicate whose W-2 it is.
Did both you and your wife live and work in New York all year?
Another possibility is that the state has information indicating that you or your wife had some other income besides your W-2s. Did either or you have any other income that you did not report?
