I had to amend my tax return because of a W2 I forgot. I printed and mailed the required forms. However, my status on turbotax still says ready to mail. Will that change?

No, it will not. Once you print a paper return for mailing, TurboTax has no way of obtaining feedback regarding next steps. The choice to sign and mail the return or not is entirely in your hands.
