After the state update failed today (2/21) I am now unable to use TT. If I uninstall TT, will it affect the work that as been done (i.e. taxes already filed)?

It is showing you are using Turbo Tax Online, which is unable to be uninstalled. So when you logged back in, all the work would still be there.

However if you do have the download version, uninstalling the program will cause you to lose everything.
