Tuition reimbursement from employer. I was taxed on $22,280 (received $5,250 tax free). Do I differentiate the money I paid on taxes somewhere other than W2?

No, you do not have to do anything extra to specify this. Only the $22,280 will be reported (and taxed) on Box 1 of your W-2.
Keep in mind if you are claiming a credit for education, only amounts above reimbursements (if any) can be claimed.
