I am a teacher CEO of a 501.C.3 organization - set up to to partner with my district on several educational initiatives. Can I deduct monetary and /or like-kind contributions to the 501.C.3 on my taxes if I do not itemize?

Maryland does not permit you to itemize deductions on the state return if you did not itemize on the federal return (page 11 at https://forms.marylandtaxes.gov/18_forms/Resident_Booklet.pdf). Sorry.
Note, however, that if for some reason you did not take the educator deduction on the federal return (line 23 on Schedule 1 (1040)), you can take the deduction on your state return. Please see code tt on page 11 of the link above.
No.
You can still deduct direct expenses you paid for supplies for your classroom, up to $250 per taxpayer. See https://www.irs.gov/newsroom/educators-can-claim-deduction-to-get-money-back-for-classroom-expenses
However, as Critter says, any contributions to a 501(c)(3) are deductible on the federal level, only if you itemize.
Note that on the state level, you MAY be able to deduct such contributions, because the Standard Deduction is still less than the federal Standard Deduction in some states.
What state do you live in?
I live in Maryland.
