On my W2 in box 14 it lists PASUI. I found out that it means Pennsylvania unemployment compensation insurance. However, I never filed for unemployment. Why would this be there and should I amend my W2?

More likely it is what you paid out to PA as an insurance premium, in case you do file for unemployment at some time in the future. The Category or description you would use, appears to be the last one. "Other - not on above list"
IF you filed your tax forms without entering it....you can probably ignore it as it appears o be an FYI telling you how much you paid into the system.....and no particular credit or deduction is involved.
I did file my taxes with it entered as it automatically generated it (Pennsylvania unemployment tax). I did not list it as Other-not listed above. Will this effect my return at all?
No, this entry will not affect your return at all. Your entry here will not affect your ability to e-file or your tax calculation.
"PASUI" is the premium paid for your state unemployment insurance coverage. As @SteamTrain pointed out before, this gets paid whether you make an unemployment claim or not.
While theTurboTax Identification of Description or Code does have the option to select "PA Unemployment Tax", "Other-Not Listed Above" is also acceptable.
Employers can put just about anything in box 14; it's a catch-all for items that don't have their own dedicated box on the W-2.
If you don't see a category in the TurboTax list that matches your W-2 entry, don’t worry. We'll figure out if it impacts your return or not. If we need more information, we'll ask you.
PASUI is a mandatory unemployment tax that is paid to the state. There are 2 parts, the employee portion that is is usually a very small amount - around .0006 times your gross pay; and the employee's portion - about 30 times your portion. I believe it just goes to help pay everyone's unemployment costs, much like social security: you're not using it right now, but your deduction helps to pay social security for those now collecting it.
