No, you can't resubmit or amend an extension request.  But, you can simply make an additional payment if the change to line 5 results in more projected tax liability.

On the tax extension, I originally submitted line 5 as $0 instead of the figure on my W2. I need to update my tax extension to reflect my W2. So there is no way in TurboTax to update my tax extension and have it resubmitted? If not, should send an updated tax extension via mail with a note on it? Thanks a ton for the help.

I am going to assume that you made a larger payment than was necessary because of that error?  Those lines on the extension form are for estimating projected liability so you can make a payment (extensions give you more time to file - but not actually more time to pay).  Since the lines are estimates, there is no mechanism to update the extension form.  You would simply file your return with the correct information.

It's not going to hurt if you want to try and resubmit it via mail (for piece of mind).  But, the likely end result is that the IRS is going to discard the note and when they go to process the extension it will get rejected because one has already been filed under your SSN.  Like I said above, the form itself isn't designed to be corrected - the normal path to correct it is to just file your tax return with the correct information.
