Had you earned four years of college credit before 2016

What do I have to reply to that question if I earned my undergraduate degree in South America, and  on 2016 I was doing my 2 year program masters in USA. I haven't studied anything else in the USA.

The purpose of this question is to determine if you qualify for the American Opportunity Credit which is only for undergraduate students in their four year of undergraduate study. Since you are a graduate student, you don't qualify for it and you answer would be yes.
