An unmarried couple lives together all year, she earned less than $4000, and he provides more than 1/2 the support. So far, so good. He can claim her as a dependent as well as their 2 children. However, she files her taxes separately and claims her son by another relationship as a dependent. Can he still claim her as a dependent?

No. A dependent cannot have a dependent. If "she" is being claimed as someone else's dependent, she is not able to claim having a dependent.
