Separate state tax

My wife is a school teacher in Florida. We have filed taxes with the state, when applicable, and with the Federal government. However, she also received payment for assisting with grading for a national school standard board based out of Missouri. Will this require us to file in Missouri as well?

No, not if she is a resident of Florida and did the work in Florida.
