Tax return shows spouse as able to be claimed as a dependent which is not true. Does this affect my refund amount?

you can not use MFS if it's indicated that your spouse can be claimed as a dependent.  as a matter of fact,  a spouse being claimed as a dependent is not permitted in most cases. so it would seem you entered something incorrectly in your spouse's personal info.
