I just noticed while starting this year's taxes, that I had mistakenly associated my wife's name to one of my W2s on last year's (2020) tax form. We file jointly, and the total income would probably not affect any of the numbers, but I'm curious if I would need to file an amendment in this case.

It could become a problem if by putting all of the income under one spouse you got a refund for paying excess Social Security. When you add up the amounts from box 4 of the W-2's you put under your own name does the amount exceed $8537.40? If so, you need to amend so that you do not get a letter from the IRS requiring you to pay back some of your 2020 refund plus penalties.
