I have to amend a return where I had purchased the Turbo Tax Audit Defense. The amendment is to fix an overstatement of dispursements. Will this affect the Audit Defense?

Amending your return will not mess anything up with your Audit Defense. Your original or amended tax return is eligible as long as you purchased PLUS Help & Support, MAX Defend & Restore, or Premium Services for TurboTax Online or Audit Defense for TurboTax CD/download when you filed your return.
