I’m trying to file form 1040-X to amend a tax return that was not previously filed with Turbotax. Do I have to enter all the information on my original tax return before selecting “amend tax return”? It was giving me some issues : not populating my original amount (column A) and line 16 (total amount paid with original return) on form 1040-X…

Yes you first need to fill out your return like it was originally filed. Then save your return. Then when you hit the Amend button the refund monitors at the top should reset to zero before you make your changes.
The amended return will only be the difference you get back or need to pay. You can confirm this by printing your amendment and looking at the 1040X. Line 18 should be your original refund amount and line 21 should be your additional refund. If you paid on your original return it will be on line 16 (but don't include any penalty on 1040 line 38). If there is an amount owed with the amendment, it will be on line 20.
You will have to print and mail the Amended return.
What year? To amend a return you did somewhere else or you don't have the original .tax file, you would first have to fill out Turbo Tax like you are doing the original return and pay for it. You have to recreate the actual wrong original return. Then when it matches each line that you really filed you have to tell Turbo Tax you will mail it so it thinks it was filed. Then you can amend it.
For the current year you can use the online version by first recreating the original return exactly as filed. Then pay the fees and say to mail it and print it out so it think you filed it. Then when it matches every line you can amend. How to Amend the current year

Thank you for your response. It is for 2023 tax year. I filed with a CPA and the taxable distribution for Roth IRA was entered incorrectly, resulting in a higher tax owed than it should have been. I already purchased the Desktop version for 2023 tax year. I saw the option “Amend a return after it was filed and accepted by IRS”. When I selected that option, it took me to the screen “What do you need to change on your return?” The federal refund was then $0. From your advice, I should recreate my originally filed 2023 tax return then hit “never mind, I don’t need to amend”? Because when I tried to file a different return, the penalty was added to the total amount owed… I don’t know a way around that.
