If I e-filed before April 15th, and then realized I need to make a change, can I re-file again before April 15th, or do I need to go through the amended return process?

No, you cannot re-file again even though it is before April 15th. In order to change to from Married Filing Separate to Married Filing Jointly, you must go through the Amendment process. See the TurboTax FAQ below for instructions:
