If the IRS did not adjust your 2020 return for you automatically ... then you need to mail in an amended return to get any refund due to you.
If you are referring to the exclusion of up to$10,200 of unemployment benefits from income for tax year 2020, it depends on when you filed your 2020 return and whether you used TurboTax. As Critter33 stated, the IRS automatically allowed the exclusion and issued refunds for taxpayers who filed their returns early in 2021 before the exclusion was granted and programmed. Check your records to determine whether you received the exclusion after filing your 2020 return.
See here for more information from the IRS on this topic. As of now, the IRS is no longer automatically calculating the exclusion.
The exclusion was available for 2020 returns only.
If you are preparing a 2020 return for the first time in TurboTax, the program will already include any exclusion.

See this article for more information on filing an amended return. The form is expected to become available later this week.
If you are instead referring to the overpayment of unemployment taxes you paid as an employer, please provide more details.
