As your spouse does not have an SSN or ITIN, you would need to apply one for her. Please see ITIN
Per IRS, you cannot e-file a tax return that does not include an SSN or ITIN. You can still use the TurboTax program to prepare your tax return and required tax forms. Print and attach them to the Form W-7 with the required documentation and submit all the papers to the IRS by mail.
Here is what you would need to do in the TurboTax program,
-As you enter the information, leave the spouse's SSN space blank
-After entering all your information, print out returns and required forms, then write APPLIED in the SSN space on the forms.

Yes. You should. Just enter her SSN number in the appropriate space and e-file the tax return.
