I paid over$4000.00 in charitable contributions but it did not seem to affect my taxes due. why is this?

Have you entered all of your other itemized deductions yet?  

TurboTax starts off giving you the standard deduction.   Once you enter enough of your total deductions (allowed medical, taxes, interest, charitable donations, deductions subject to the 2% floor, etc.) that it exceeds your standard deduction, then TurboTax will switch to itemized deductions on Schedule A.   Medical is further limited; only the amount over 10% of AGI (7.5% for age 65 or older) will count--and only if you can itemize deductions.

Have you entered enough total deductions for TurboTax to switch from standard deduction to itemized deductions?  If you haven't entered all your deductions yet, wait until you've entered the rest before deciding how TurboTax is handling them.   Until the total surpasses the standard deduction, they won't start to count on the Federal side.
