If I re-import does it replace (completely overlay) the prior data?

How do I delete a prior import?  Do I have to delete each line item individually on the "Review All Your Charities" screen (TT CD version)?

You should delete the first import and import the new info ... do not try to import over a prior import as dumb things can happen.
