2020 amended form and 2021 filing

Do I start my 2021 filing using my 2020 amended file?

Yes (assuming that you actually filed the amended return with the IRS). If you filed an amended return (Form 1040-X) for 2020, when you start your 2021 tax return, transfer information from your amended 2020 tax return.

Yes ... you always use the most recent return filed.  However when you are asked for the 2020 AGI you must use the figure from the original return so save a PDF of the return first.
