Can I get a tax deduction for personal HSA contributions made after federal taxes (not through payroll deduction)?

All of my personal HSA contributions are made by me (not through my employer) from my post-tax income. Am I able to deduct these contributions (up to current limits) when I file my taxes?

Yes.  Such contributions show up on line 25 - "Health savings account deduction. Attach Form 8889" - of Form 1040, reducing your Adjusted Gross Income.

But the constraint is that you have to be on a high deductible plan for the year in order to take advantage of this write off?

Not just "high deductible", it must be an "HSA-qualified HDHP"

How do I file on turbo tax if my w2 does not reflect the HSA contribution I made outside of my paycheck, directly to my HSA account? Can I subtract the amount from my AGI?

When you complete the HSA section of TurboTax, TurboTax will ask how much you personally contributed.  When you make this entry, TurboTax will include this amount on Form 8889 line 1 to be used in calculating the deduction that will appear on Schedule 1 line 12 that reduces your AGI.
