I own an airbnb / vacation rental in another state, I don't have any other income from that state other than the airbnb vacation rental. Do I need to file an income tax return for the state the air bnb / vacation rental is located in?

That would depend upon the state in which the rental is located? Which state?
If the state levies an individual income tax, then you have state source income from that state and likely need to file a return.
Generally, yes, if you meet the filing requirements for the state your AirBNB is located in. Please see Why would I have to file a nonresident state return? and contact your state to see if you're required to file. See How do I contact my state Department of Revenue?
What if the rental property is in Puerto Rico but main residence is in the states ? Airbnb issues a 1099-k form am I required to file taxes there or in the states?
Since the rental income is from sources within Puerto Rico and you reside within the states, you would need to file taxes to report the rental income within Puerto Rico per IRS Publication 570.
