Box 14 on W-2 states "Section 125 4256.20". Is 4256.20 the $ amount to be entered?

Yes.  It is related to your employer benefits plan (a "cafeteria plan"), but it is for informational purposes only.  It does not have an effect on your tax return because the amount has already been excluded from your Box 1 wages.
