My spouse is not available to sign the tax return and we are filing jointly. Can I sign on her behalf?

When you e-file there is no physical signature on the tax form.   You "sign" electronically by entering your AGI or PIN.   If your spouse agrees to a joint return and wants you to enter that for her, it's fine---you do not have to get her to click the mouse or get on the keyboard for that.   If you are paper filing, then, well....yes she should sign her own signature on the line that says "Sign here."
