The short answer is "No", your return has to be filed on paper. From the AARP Tax-Aide "Duckbook":
For MFS returns, if in scope, where the spouse’s SSN or ITIN is not known, enter 111-00-1111. The return
cannot be e-filed; the return must be paper-filed; write "unknown" wherever the spouse’s SSN appears on
Form 1040 and related forms.
(The 111-00-1111 may be specific to TaxSlayer. I don't know offhand what TurboTax uses.)
A longer answer arises if you live in a community property state such as CA (see
for the full list). In that case, you are required to divvy up income into individual versus shared. In your instance, you may qualify to just report your own income. The requirements for this are listed on pages 8-9 of IRS publication 555 ( https://www.irs.gov/pub/irs-pdf/p555.pdf ) and basically say that if you lived apart all year and have no intention of reconciling you can set the spouse's income to zero.
