For the calculation of square footage in the simplified home office deduction, should I include the square footage of a closet?

If the closet is used exclusively as part of your home office then it should be included with the square footage of the office. If it is used for personal purposes, then don't include it.

Yes, but only if it is used strictly for business. If you have personal items in it, it is not business use.
