I live in Michigan, but had winnings from Draftkings which has a Massachusetts address on the W2. However, all of my earnings were from online, never physically being in Massachusetts. Do I have to file state taxes in Massachusetts?

No, you do not have to file a Massachusetts tax return, unless they withheld Massachusetts taxes in Box 15 of your W-2G which is very unlikely. If they did, you would have to file a return just to claim a refund of the taxes.
In turbotax, when it asks if I earned income (including gambling winnings) in another state, do I say "yes" or "no"? It seems like when I say "yes" it wants to start a Massachusetts state tax form.
You would say No. This income is considered earned in Michigan since you were not physically present in Massachusetts when you won. The location of the company does not change that.
