If the only change in your wife's W-2 was a "corrected" amount for Box 12, Code DD, then the answer to your question is no, you do not need to amend your tax return for that item only.
Code DD is a requirement of the Affordable Care Act (a.k.a. Obamacare), whereby employers report the amount of pre-tax health insurance premiums that were paid by the employer, on behalf of the employee, during the calendar year. But it has absolutely no actual effect on an individual taxpayer's income tax return. It is intended purely for informational purposes only. In fact, you can even omit a Box 12 DD entry entirely, and it won't affect processing of your tax return (or refund, if applicable).
