When I transfer last year's return, do I use the original return or the amended return?

I amended my 2020 return.  If I use the amended file to transfer data for my new 2021 return, does it contain all the information needed to be transferred?  If I use the original file, might it miss something that is needed to be transferred for 2021?

Yes transfer from the Amended file.   It will have everything.

Correct ... transfer the last filed return however you will need the AGI from the original 2020  return to efile the 2021 return.  The AGI on the amended return will not work.
