I expect my spouse & I will have an AGI of less than $28,000 annual & if I have a charitable deduction of $1,000 will that make a big difference in my tax return?

Charitable contributions are an itemized deduction on Schedule A.  The total of all your itemized deductions would have to be greater than the Standard Deduction for your filing status to have any affect on your tax return.

 

2022 Standard Deduction for Married Filing Jointly (under age 65 and not blind) - $25,900

@pdac1 - further, in 2021, you could reduce your AGI by up to $600 with CASH charitable contributions.  That will not be the case in 2022 (unless Congress acts), so the charitable contribution is unlikely to have any impact.
