Filing Married Separate, I can claim the exemption for my spouse if they have 0 income, not filing joint & not claimed by another taxpayer. How do I enter it in TurboTax?

Yes.  In the Personal Info section, select Married Filing Separately.  As a follow up question, you will see "Let's see if you can claim _____'s personal exemption."   Check the first three boxes (see attachment).
