Do I need to give a handwritten signature if I have e-filed with e-signature? Both have been accepted by state and federal.

No, you do not need a physical signature.  By e-filing, your 'electronic' signature is sufficient.  When you e-file, you do not have anything to physically mail to the IRS.  Just save all of your paperwork for your own records, including a printed copy of your return.
