Can I file my taxes prior to a ROTH 401k over contribution being withdrawn?

My wife had two employers in 2020 and had an excess contribution to her ROTH 401k for the year as a result. We have been trying to get her current employer to back out contributions and it has not been a fast process.

My question is, can we file our 2020 taxes prior to her current employer removing the excess or do we need to wait for a revised 2020 W-2? I haven't seen a direct answer to this question, but from what I gather it should be acceptable to file our taxes because any income/gains from the excess contributions will be taxed in 2021 income.

Yes, you can file with the correct information if you do not want to wait for the W-2C.  The only consideration is to compare your return with the correction when it comes.  If it does not agree with the return you filed, then you would need to amend your original return.
