We are building a barn on a piece of land, no house yet. office space will be included in the structure. can i deduct a portion of the cost to building the barn?

No, oyu cannot deduct it.  It would be capitalized as an asset once the business is open and running.  Costs that would normally be capitalized as assets cannot be start-up costs
