The IRS has sent me a letter asking to substantiate line 25b federal taxes withheld from Forms 1099R.
I have reviewed my Turbotax filing, checked my entries, and cannot substantiate how TurboTax calculated the total of my 1099R entries. The error is almost a factor of 2 more than I entered in the program!
Any thoughts? I see another person reported a same error, but subsequently reported that he found the error.

Line 25b is not calculated - it is simply an addiction of all 1099 box 4 tax withholding that you entered. This is almost always caused by downloading the 1099-R's from a financial institution more than once or also manually entering them.
Return to your account and print all forms and worksheets "Tax payments worksheet" lines 12 & 13 for the withholding entered.
Save a PDF of the return with the worksheets to see how that was complied. And/or open the program and review your entries ... maybe you entered something in the wrong place and/or twice.
NOTE: When you open your return back up, do not make any changes in it if it's already been filed. If you later have to amend the return, it has to start off exactly as it was when it was originally filed.
NOTE: Remember all PDF tax documents and tax data files are very sensitive files, since they contain your personal ID info, financial data, and possibly bank account numbers, etc. Be sure to store them safely and securely to guard against computer theft, hacking, etc.
