I received a state of MA notice of intent to assess for 2014. They are right. I owe $2431. Will I later have state int./penalties and have to file amended 2014 Fed Tax

They are right. I file jointly. I input my wife's Fed. Tax witholding in the place for state witholding. I have until 1/12/2018 to pay the difference-$2431.

If you made that error on your federal tax return, if you did, you will need to amend your Federal return (1040) so the information provided is correct. If you did not make that error on your federal return, there is no need to file an amended 1040 (Federal return).
