I'm amending 2018 tax return status to married filing separately since my wife is non-US citizen and she doesn't have a SSN. On 1040x and 540x, is her signature required?

You have a problem and need to seek local paid tax help.  You are not allowed to amend a joint return to married filing separately after the filing deadline.  Hopefully a pro can help you sort out what to do.
