I live in Kentucky but work in Ohio. I filed taxes for Ohio and Kentucky. After filing (and paying Ohio $1,700) I found out that I did not need to file for Ohio because OH and KY have a reciprocal agreement. Was told to file an amended return. I no longer have access to the machine I used TurboTax Desktop with. What do I do?
No, you cannot amend from TT online. You have to have your original tax file.
You would have to re-enter your original tax file. Select file by mail (but don't actually print and mail). Then you can amend.
Before contacting TT support, you may want to try preparing an amended return by hand. Even if you fail, you'll be bettre prepared to talk to support.

You are not required to file a local city return, but you are also not entitled to a refund of the city withholding (box19 on your W-2), unless there was an error in the amount. If you are subject to a KY county or city tax, you should check to see if the KY city will give you a credit for the OH city tax withheld. KY does allow an itemized deduction (Schedule A) for local tax paid.
