Yes, if your efile is Accepted you have to wait for the original return to be fully processed and you get the first refund (or pay the tax due). Then you can amend. Amended returns will take 4 months or more to process. If your efile is rejected you can just fix it and re efile.
Why are you amending? As asked on your other post you can't change the filing status if you filed a Joint return.
