I want to pay my taxes. I am confused about the ACH# and the regular branch routing #

Turbo tax shows to use my bank routing #, unless I have an ACH #.  My check has an ACH # but it is not located on the check where Turbo Tax says.  Does it really matter which routing # I use.   I direct pay a lot but I don't use an ACH #
