No, you can't claim her as a dependent. You would want to file a joint return with her- she doesn't need income to file a return with you. (I am assuming she is not the qualifying dependent of someone like a parent)
Your spouse is either your joint return spouse or you file separately. They are never listed on the return as a dependent.
