Assuming that you are a US person ( citizen/GreenCard/ Resident for Tax purposes), yes you can file a 1040 with a status of Married Filing Separate. Note though that you still have to provide details of your spouse ( SSN, Name etc ) and will be limited to using the same type of deduction as that of the spouse ( Standard or Itemized )..
Generally MFJ is often more tax beneficial compared to MFS. If you have dependents for whom you maintain a household, you can also file as Head of Household
