my federal tax due is being deducted from my checking account. Will this include my 1st quarterly estimated tax payment or do I need to mail check for 1st estimated federal tax payment?

No, it won't be included.  You will have to mail a check or pay the estimate online at the IRS website.  Here is the link https://www.irs.gov/payments
