I mailed my Form 3520 to the IRS PO Box using certified mail, but the tracking still shows “moving through the network” with no delivery confirmation. I’ve already submitted a missing mail request with USPS. If delivery can’t be confirmed, should I resend the Form 3520 before April 15 with a cover letter marked “possible Duplicate Filing” and send it via FedEx to a physical address instead of the PO Box?

The instructions for Form 3520 state expressly to send it to this PO Box - don't use another address:

A duplicate filing of Form 3520 may accrue an automatic civil penalty onto your account, which could drastically slow down processing of the original return. This might be a question for an attorney.
If you have the certified mail receipt, you should be able to furnish this as proof of timely filing if the form is truly lost. Make a copy of your missing mail request, and keep that with your records as well. I thought everything I sent last year by certified USPS was lost or even tossed away; I didn't get a single confirmation of delivery, so I wouldn't be so quick to assume it's truly lost in the mail. All the certified mail I sent was delivered, eventually.

To see if the form has posted to your account, you can also send Form 4506-T Request for Transcript and put "3520" on Line 6.
THANKS! i am hoping USPS delivers it soon. If not, I will of file form 4506-Tin April and putting “3520” on Line 6 of return of transcript.
