It depends. However, if your employer did not offer health insurance, you would not qualify for a healthcare penalty exemption for that reason alone.

The penalty (shared responsibility payment) should be reported on your federal tax return on Form 1040 Line 61/Form 1040A Line 38/Form 1040EZ Line 11 Health care: individual responsibility, so you should not need to send additional payment.
