No, do not make any changes to your W-2; enter that as you see it. To enter quarterly tax payments that you made in 2019, follow these instructions.
If you made estimated tax payments in 2019 towards your federal, state, or local taxes, enter them in the Estimated and Other Income Taxes Paid section.

No...Your withholding total payments that are on your W-2 are entered only when you enter all your W-2 information onto the software W-2 form....nowhere else
Quarterly Estimates Federal (or State) payments are those "extra" payments you arranged on your own to pay quarterly....outside of work.
People who are self employed...or who happen to have a lot of extra outside investment income, usually use estimated payments to avoid a huge tax bill when they file (And possible penalties for underpaying during the year) . Unless you made those extra payments yourself, you wouldn't have anything to enter for Quarterly estimates.
