Yes, you can file as Married Filing Jointly on the 2021 tax return if your spouse passed away in 2021.
When entering your spouse's information in the My Info section of the online program there will be a section which asks if they have passed away. Select this and then enter the date of passing in 2021.

You may be unable to e-file. In the past, it was not clear if this was a security precaution (your spouse's SSN may be blocked to prevent fraud) or a Turbotax glitch, since some people reported being able to e-file with other tax companies. If you are rejected from e-filing, you can print, sign and mail your return.
